Mutations in the RpoB Gene of Multidrug-Resistant Mycobacterium Tuberculosis Isolates from Semarang, Indonesia
Authors
Abstract:
ABSTRACTTuberculosis is an infectious disease which occurs widely in the world and becomes one of the top 10 causes of death worldwide. Mutations of the RpoB gene cause the resistance of Mycobacterium tuberculosis to rifampicin that contributes to the occurrence of MDR-TB. This study aimed to determine the pattern of rpoB gene polymorphisms in the MDR M. tuberculosis in Semarang, Indonesia. Most mutations occurred at codon Ser531Leu (13 isolates), codon Gln490Lys (7 isolates) and codon 526 (5 isolates). Variations of mutation mostly occurred at codon 526 with 4 variations of nucleotide changes, including His490Leu, His490Cys, His490Ser and His490Tyr. The types of mutation were missense mutations (30 mutations) and silent mutation (1 mutation). The mutations in the rpoB gene of MDR M. tuberculosis isolates in Semarang showed genetic variations that caused high levels of resistance to rifampicin. Keywords: MDR M. Tuberculosis; rpoB gene mutations; rifampicin resistance

Mutations in the 81-bp rifampin resistance-determining region (RRDR) of the rpoB gene were analyzed by DNA sequencing of 50 Mycobacterium tuberculosis clinical isolates (44 resistant and 6 sensitive) from various parts of India. Fifty-three mutations of 18 different kinds, 17 point mutations and one deletion, were observed in 43 of 44 resistant isolates. Three novel mutations and three new alle...

Mutations in a 69-bp region of the rpoB gene associated with rifampin resistance (Rif(r)) in 100 isolates (82 Rif(r)) from three states of Brazil were studied. Twenty-one different kinds of mutations were identified in the Rif(r) isolates, and six new alleles are described.

Mutations in the 81-bp rifampin resistance determining region (RRDR) and mutation V176F locating at the beginning of the ropB gene were analyzed by DNA sequencing of 86 Mycobacterium tuberculosis clinical isolates (72 resistant and 14 sensitive) from different parts of China. Sixty-five mutations of 22 distinct kinds, 21 point mutations, and 1 insertion were found in 65 of 72 resistant isolates...

In many developing countries and outside hospital settings, the characteristics of endemic Mycobacterium tuberculosis strains resistant to multiple drugs remain unknown. In a community-based referral and therapy program in northern Lima, Peru, beginning in 1996, patients found to be failures on standard regimens were referred for drug-susceptibility testing of their isolates, and those found to...

Objective: The Rifampicin resistance and susceptibility of Mycobacterium tuberculosis are caused by mutations in the 81-base pair region of the rpoB gene encoding the b-subunit of RNA polymerase. Methods: Isoniazid resistance of M. tuberculosis is related to mutations in inha , oxyR and ahpC genes which 30 to 90 percent of Isoniazid resistance is occurred in 3015 codons of kat...
